In the realm of high fashion, a series of bizarre events unfolds, commencing with a high-speed motorcycle chase through the streets of Rome, where two assassins are hot on the heels of pop sensation Justin Bieber. The singer manages to take down one of his pursuers before seeking refuge at the residence of legendary musician Sting. However, his sanctuary is short-lived, as he is cornered by the remaining assassin and meets a grisly end. With his final breath, Bieber takes a selfie, striking a pose reminiscent of a iconic modeling expression, and shares it online.
This peculiar incident is merely the latest in a string of similar events, where various celebrities, including Demi Lovato, Usher, Lenny Kravitz, and Ninja from Die Antwoord, have all fallen victim to the same fate, each leaving behind a selfie with the same distinctive pose. The Fashion Division at Interpol takes notice of this trend and recognizes that the key to unraveling the mystery lies with a former modeling icon, Derek Zoolander.
A flashback sequence reveals the tumultuous life of Derek Zoolander, once the epitome of male modeling, whose career and personal life began to unravel 15 years prior. The collapse of his charitable organization, The Derek Zoolander Center for Kids Who Can't Read Good, resulted in the tragic loss of his wife, Matilda, and left his close friend and fellow model, Hansel, disfigured. Derek's subsequent descent into madness led to him losing custody of his son, Derek Jr., and his decision to abandon the world of modeling to live a reclusive life as a "hermit crab" in rural New Jersey.
Derek's solitude is disrupted by a visit from Billy Zane, who extends an invitation from the enigmatic fashion designer, Alexanya Atoz, to participate in a high-profile fashion show in Rome. Derek is initially hesitant, but the prospect of reuniting with his son and regaining his former glory eventually wins him over. Meanwhile, Hansel is living in the "Uncharted Malibu Territories" with a group of strangers, whom he refers to as his "orgy." However, upon discovering that all the members of his orgy are pregnant, Hansel flees, only to be reunited with Derek in Rome.
The two former models are escorted to the fashion show by an androgynous child, VIP, and are introduced to the eccentric Don Atari and the androgynous fashion icon, All. The fashion show descends into chaos, with Derek and Hansel being publicly humiliated and doused in prunes. The ordeal comes to an end when they are approached by Agent Valentina Valencia, who requests their assistance in unraveling the mystery behind the series of celebrity murders.
At Interpol, Valentina presents Derek and Hansel with the evidence, including the selfies of the deceased celebrities, all of which feature the same distinctive pose. Derek recognizes the expression as a variation of his own signature look and recalls a bizarre advertisement he once appeared in, featuring a half-man, half-cow creature. The investigation leads them to an orphanage in Rome, where Derek's son, Derek Jr., is residing. However, upon meeting his son, Derek is taken aback by the boy's unexpected appearance.
As the story unfolds, Derek and Hansel become embroiled in a complex web of intrigue, involving the villainous Jacobim Mugatu, who is secretly manipulating events from behind the scenes. Mugatu's plan is eventually revealed, and Derek, Hansel, and Valentina must join forces to thwart his evil scheme, which involves sacrificing Derek's son to gain eternal youth and beauty.
The thrilling conclusion features an action-packed showdown between the heroes and Mugatu's accomplices, including Alexanya Atoz, who is revealed to be Mugatu's partner in crime, Katinka. The battle culminates in a dramatic confrontation, where Derek, Hansel, and their allies must use their unique skills to defeat the villains and save the day.
